In situ hybridization, as the name suggests, is a method of localizing and detecting specific mRNA sequences in morphologically preserved tissues sections or cell preparations by hybridizing the complementary strand of a nucleotide probe to the sequence of interest. In situ hybridization is widely used to visualize transcribed sequences in embryos, tissues, and cells. For whole mount detection of mRNAs in embryos, hybridization with an antisense RNA probe is In situ hybridization (ISH) targets specific nucleic acids within fixed tissues and cells to indicate a gene’s presence or expression while allowing simultaneous visualization of the tissue’s morphology. ISH staining is not prone to fading as hybridization signals are in fluorescent in situ hybridization (FISH).

The nucleic acid sequence is bound specifically in a tissue section by complementary base pairing, that is, hybridization, with a detectable nucleic acid segment called a probe. In situ hybridization (ISH) is a powerful technique for localizing specific nucleic acid targets within fixed tissues and cells, allowing you to obtain temporal and spatial information about gene expression and genetic loci.
